Synthesis and evaluation of acetaminophen derivatives as analgesic, antipyretic and anti-inflammatory agents

In the present study, Dicarboxylic acid bis (4-acetylaminophenyl) ester (AP1-AP4) and (4-acetylaminophenoxy) acetic acid (AP-5) have been synthesized by the reaction of acetaminophen with dicarboxylic acid chlorides and chloroacetic acid, respectively. The esterification of (4-acetylaminophenoxy) acetic acid with different alcohols yielded rest of the compounds (AP6-AP13). The characterization of the compounds was done by IR, 1H NMR and elemental analysis. All the synthesized compounds were screened in vivo for analgesic, antipyretic and antiinflammatory activities. Appreciable activities were observed for many compounds.
