High performance liquid chromatography with PDA det ector for combined determination of hydrochlorothiazide, amlodipine an d valsartan

A simple, precise, and accurate HPLC method for sim ultaneous estimation of hydrochlorothiazide, amlodi pine and valsartan as the bulk drug and in pharmaceutical do sage forms. Chromatographic separation of the drugs was performed on Zorbax C18 (250 x 4.6 mm; 5 μ m particl e size) analytical column as the stationary phase. The solv ent system consisted of 0.1M NaH 2 PO 4 and methanol in the ratio of 60:40 (v/v) as mobile phase. Evaluation of the separated drugs was performed using a PDA detector covering the range of 200-400 nm. All the three dru gs were resolved with the retention time of 3.624 min, 5.48 4 min and 14.943 min for hydrochlorothiazide, amlod ipine and valsartan, respectively. The method was validated w ith respect to linearity, sensitivity, precision, a ccuracy and robustness in accordance with ICH guidelines. The v alidated method was successfully applied to the com mercially available pharmaceutical dosage form, yielding good and reproducible result.
